Abstract: The cited analytes were extracted from alkaline plasma (0.5M-NaOH added) into cyclohexane - ethyl acetate (1:1) and transferred after solvent evaporation into mobile phase for HPLC injection. The HPLC column (10 cm .times. 4.6 mm) contained Hypersil Phenyl (3 .mu.m), the mobile phase (1.5 ml min.minus.1) was 0.02M-NH4 formate buffer (pH 2.75) - acetonitrile (3:7) and detection was at 240 nm. The calibration range was 1 to 500 ng ml.minus.1 for tacrine and three of its monohydroxy-derivatives, measured vs. 1,2,3,4-tetrahydro-N-methoxyacridin-9-imine as internal standard. The coeff. of variation (n = 6) were .ltoreq.10% at 5, 50 and 500 ng ml.minus.1 with recoveries of tacrine of 92 to 96% and 67 to 72, 32 to 35 and 83 to 86% of the 1-, 2- and 4-hydroxy derivatives, respectively.
